明反応の産物を介した高CO2応答ネットワークの解明

Research Abstract

シロイヌナズナ野生株およびpgr5変異株を高CO2濃度に馴化させ、大気CO2濃度に戻した後豆5分、30分、60分、120分に発現が変動する遺伝子を調べた。また大気CO2濃度に馴化した植物での遺伝子発現も調べた。独立のアレイ実験で変動が確認された遺伝子のうち、din2の発現は、大気CO2濃度に移した後120分で誘導された。また大気CO2濃度に馴化した植物でも高い発現が見られた。一方、pgr5では常にdin2の高発現が確認され、特に大気CO2濃度に移した後30分後に大きな発現誘導が見られた。din2はβ-glucronidaseをコードし、暗処理、糖欠乏で発現が誘導される。このことは、野生株では高CO2濃度において糖の蓄積が起きる一方で、pgr5では糖の蓄積は起こらないことを示唆している。このことは、サイクリック電子伝達が糖の蓄積を介して高CO2濃度の認識に関与している可能性を示している。
また昨年度に引き続き、サイクリック電子伝達を特異的に阻害する薬剤の解析を行なった。AAL1と名付けた薬剤は、PGR5依存のサイクリック電子伝達を特異的に阻害し、またその作用機作はアンチマイシンAと同じであることが示唆された。一方、アンチマイシンAと異なる部位を阻害する可能性のあるAAL2を選抜した。

Reason

野生株とpgr5変異株でCO2濃度の変化に異なるレスポンスを示す遺伝子を探索していたが、安定した結果を示す遺伝子を見いだすのに予想以上に時間を要した。最終的に複数のアレイの結果を比較することで候補遺伝子を得た。当初の計画外であったが、PGR5異存のサイクリック電子伝達を低濃度で特異的に阻害する薬剤を得ることに成功した。

Strategy for Future Research Activity

今後は、糖を介さない高CO2濃度の情報伝達に関わる遺伝子の特定が望まれる。またサイクリック電子伝達阻害剤は、作用機作の特定が、次の重要ステップになる。
