Research Abstract

生活習慣病における代謝異常の中でも、脂肪酸の代謝異常が生活習慣病の発症と悪化に深く関わっている。我々は炭素数12~16の飽和および一価不飽和脂肪酸を基質とする脂肪酸伸長酵素Elovl6をクローニングし、Elovl6欠損マウスの解析から、本酵素による脂肪酸組成の変化が肝臓におけるエネルギー代謝やインスリン感受性に重要であることを明らかにした。近年、代謝性疾患の基盤病態として全身の非感染性の慢性炎症が注目されているが、本研究では、Elovl6によって制御される細胞内脂肪酸組成の変化と自然炎症の関連を、特に生活習慣病の肝の表現型である非アルコール性脂肪性肝炎(NASH)に着目して解析した。野生型およびElovl6欠損マウスに高脂肪・高コレステロール食を負荷すると、野生型マウスでは肝の脂肪化と炎症・酸化ストレス・線維化が引き起こされるが、Elovl6欠損マウスでは肝の脂肪化は起こるものの炎症が抑制され、NASH病態が改善された。一方、アデノウイルスやトランスジーンによりマウス肝臓でElovl6を過剰発現させると、脂質の蓄積による肝臓の肥大、強い炎症性細胞浸潤と炎症の亢進、肝障害が引き起こされた。また、正常に比べてNASH患者の肝臓ではELOVL6発現が2倍以上増加していた。したがって、マウスおよびヒトにおいて、肝臓におけるElovl6の活性化は肝の炎症を促進し、NASHの発症・進展に寄与することが明らかとなった。初代肝細胞培養系を用いた解析から、Elovl6活性の亢進が肝細胞のインフラマソームを活性化し炎症を促進することが明らかとなった。Elovl6による脂肪酸組成変化とインフラマソームの関連を引き続き解析することで、生体内において炎症を制御する脂質やその代謝物、新たな炎症制御系の同定が期待される。
