Research Abstract

本研究課題は、X線1分子計測法において溶液pHを観測中にジャンプさせ、イオンチャネルの構造変化の遷移過程を連続的に計測することであった。
昨年度から今年度初めまでヨーロッパ放射光施設(ESRF,フランス)に滞在して実験系の構築・確認・データの蓄積に取り組んだ。
レーザー光を用いて観測中に溶液pHを7から4へとジャンプさせる実験系の構築に成功した。現地の研究者である、Stetten, V.D., Royant,A. 両博士と協力してレーザー光の照射輝度と照射時間、pHの変化量に関する基礎データを蓄積し、照射条件を決定した。またGuerin, L.博士と協力してX線照射位置とレーザー光の照射位置を重ね合わせ、X線照射とレーザー光照射、カメラのスタートのトリガーインタープレイシステムを構築し、観測中にpHを任意のタイミングでジャンプさせることが可能になった。
これらのシステムと研究代表者が導入した高速観測システムの連携により、閉状態から開状態への状態遷移過程を連続的に5000フレーム/秒の速度で観測することに成功した。また、自動計測システムの導入も行い、効率的にデータを蓄積することが可能になった。
帰国後、データ解析プログラムを研究代表者自身が開発し、画像データから輝点座標の時系列データを抽出し軌跡を追跡することに成功した。多量の画像データの初期データ解析を終え、結果をアメリカ生物物理学会で発表した。発表はインパクトの高い発表として口頭発表課題に選出された。
