膜透過ペプチドと対イオンを用いた準安定状態を探るセンサー分子の効率的な細胞内導入

Research Abstract

近年、HIV-1 Tat (48-60)ペプチドをはじめとした膜透過性アルギニンペプチドが細胞内へ効率的に移行する性質を利用し、これらをキャリアーとして細胞内へのタンパク質や薬物を導入する手法が広く利用されている。我々は、さらに疎水性の対イオンを共存させることでアルギニンペプチドの細胞内移行量が数倍から数十倍に向上することを見出しているが、本手法におけるアルギニンペプチドの細胞内移行機序に関しては不明な部分が多い。そこで本研究では、アルギニンペプチドの膜への集積と直接透過に寄与する細胞膜側の要因や、ペプチドとの相互作用様式について、モデル膜 (LUV、GUV)と分光学的手法を用いて検討した。
酸性脂質を含まないGUVにおいて、疎水性対アニオンのピレンブチレート(PyB)の添加の有無にかかわらず、蛍光標識したオクタアルギニンペプチドのGUV膜への顕著な集積及びGUV内移行は観察されなかった。これに対し、DOPSを含むDOPC/DOPS/SM/Chol (1:1:2:1)のGUVでは、PyBを添加した場合、速やかなGUV膜への吸着とベシクル内へのペプチドの移行が観察された。次にアルギニンペプチドの蛍光異方性減衰について、BODIPY標識したR8ペプチドの回転相関時間を調べた結果、DOPC/DOPS (4:1)LUVの添加によって回転相関時間が明らかに上昇した。また、PyBを添加することで、長い回転相関時間を示す成分が出現し、平均回転相関時間が増加することが初めて明らかとなった。これらの結果は、ペプチドの集積が起きやすい負電荷の脂質膜表面において、PyBとペプチドが強く相互作用することを示唆する。PyBそのものの膜障害性は低いものの、脂質組成特異的に膜流動化を起こすことも示唆され、生細胞において、PyBのこのような性質がペプチドの直接膜透過に寄与している可能性が考えられる。
