Research Abstract

タンパク質の複合体形成においては、様々な結合様式があることが構造解析や熱力学測定について明らかになりつつある。特に近年、複数の結合様式を持つタンパク質複合体形成過程が注目を集めている。ミトコンドリアTom20とシグナル配列との複合体形成においてはX線結晶構造解析や核磁気共鳴の実験から、少なくとも3つの安定構造が存在することが示された。我々は分子動力学計算を用いて、結晶構造間の転移や各安定構造の自由エネルギーなどを評価した。その結果、結晶構造の一つであるY状態は、結晶中で二量体を形成することで、溶液中よりも安定化していることが明らかになった。一方、A状態やM状態に関してはTom20複合体の熱揺らぎの範囲内で転移しており、実験で得られた複数の結合様式が溶液中でも存在することが明らかになった。Tom20の生物学的機能としては、特定のシグナル配列のみを選択し、膜輸送体であるTom40へ受け渡すことがあげられる。すなわち、「キャッチ アンド リリース」を効率よく行うための分子機構として、より安定な複合体(A状態とM状態)、そして、やや不安定な複合体(Y状態)を含む複数の結合様式が用意されているのではないだろうか。このような複数の結合様式を実現するため、Tom20においては2つの結合ポケットが用意されていた。一方、シグナル配列においては3つの疎水性アミノ酸残基が用意されており、そのうち2つの疎水性アミノ酸残基が各状態での分子認識に直接関わっていた。分子動力学計算を行うことで、このような相互作用におけるフラストレーションが複数の複合体構造を生み出していることが明らかになった。Tom20以外にも、新学術領域研究の班員である高木淳一(阪大)らとの共同研究によりインテグリンの構造ダイナミクスに関する分子動力学計算を行い、イオン結合とインテグリンのダイナミクスの関係を解明した。
