Outline of Research at the Start

メカノクロミック素材は、外部から力を加えることで色を変えたり発光したりする材料のことである。その中でもポリジアセチレンは比較的高感度であり、抗体を付加することでウィルスなどの抗原を色によって検出する安価なコロナ検査薬などが開発されている。本研究では、ポリジアセチレンがどの向きに、どの程度の力を加えることでどれだけ発光するのかという「力と発光の相関」が水分子の存在でどのように変化するのかを、近年自身で開発したナノ摩擦力顕微鏡を導入し世界で初めて解明する。ポリジアセチレンの水溶液中での特性を定量的にナノスケールで解き明かすことで、バイオセンサ感度向上を前進させる意義がある。

Outline of Annual Research Achievements

メカノクロミックポリマー、ポリジアセチレンがどの向きに、どの程度の力を加えることでどれだけ発光するのかという「力と発光の相関」が水分子の存在でどのように変化するのかを、近年自身で開発したナノ摩擦力顕微鏡を導入し世界で初めて解明した。ポリジアセチレンは比較的高感度であり、抗体を付加することでウィルスなどの抗原を色によって検出する、現存のものより安価なコロナ検査薬などバイオセンサ開発が進められている。本研究で得られる「水中でどのようなタイプの力に特に反応するのか?」という情報を元に、ポリマーに組み込むレセプターの位置や角度をコントロールすることで、検出物質の付着が発光を効率良く誘起させるようにセンサをデザインし、メカノクロミック・バイオセンサ感度向上を前進させる意義がある。本研究の鍵となるのが近年私の研究室で精度を向上した、ナノ摩擦力顕微鏡という基盤に対して水平・垂直両方の力をナノスケール制御する手法である。ナノ摩擦力顕微鏡と蛍光顕微鏡のダブル・セットアップを水中で稼働させ、ポリジアセチレンの「力と発光の相関」を定量的、異方的、ナノスケール、水溶液中で解明した。イオン濃度やpHを変化させながら「力と発光の相関」を測定することで、ポリジアセチレンの力感度がイオン濃度・pHによって制御できることを発見した。メカノクロミックポリマーを初めて水圏機能材料という視点で捉えることで、その感度・選択性を水によってコントロールするという新たな発想にもとづき、エンジニアリングの可能性を拡張した。
