Outline of Research at the Start

本研究では、生体高分子の形態不安定性を活用した水圏機能材料を作製するとともに、その形態を制御する因子の解明を目指す。我々がこれまでに取り組んできた骨格タンパク質、多糖、核酸などの生体高分子に関する背景(Okeyoshi K, et al: Sci Rep 2015, 5, 9581; Biomacromolecules 2016, 17, 2096; Small 2020, 16, 2001993)を活かし、普遍的な形態制御法を構築する。外部環境の変化に対する「自己集合/分解」の可逆的な形態変化には、生体高分子が持つ階層性の優位点であり、アクチュエータやDDS設計指針に重要である。

Outline of Annual Research Achievements

生体組織は水圏環境で得た分子構造から自己組織化を通して、光合成や呼吸など高次な機能を発現させている。様々な生体高分子が人工軟骨や細胞足場など医療用材料へ応用研究が加速する一方、その形態制御に基づいた材料化が発展途上にある。例えば、水に分散させた多糖を乾燥させて得た膜を水蒸気駆動型のアクチュエータに展開可能なものの、水中/空気中における形態や構造変化は未解明な点が多い。そこで本研究では、水圏環境で生まれた生体高分子、特に多糖の環境適応能力に着目して、その配向化を制御した。さらに、湿度や塩の種類に適応して形態変化する多糖膜の機能展開を目指し、領域内の連携を通して分子/ナノ/マイクロ構造を解析した。我々のオリジナル手法である界面分割法で得た高分子膜について、SAXS, WAXS, IR, 各種顕微鏡法を用いてナノメータースケールの分子配向について解析し、その異方構造を考察した。多糖は水圏環境の塩濃度やpHによって形態をダイナミックに変化させる特性を活用し、多糖数種に対して外部刺激への異方応答材料を作製した。これらは、既往材料として合成高分子で展開される刺激応答高分子などに代替する環境適応材料としても期待される。本年度において国際学会、および国内学会の発表10件以上、査読付学術論文2報を通し、活発な議論を進めた。これら多角的なアプローチを通して、水圏機能材料に関わる新学術の推進に重要なステップを踏んだ。
