Outline of Research at the Start

本研究では、非ゲノム情報であるG4/RNA-DNA ハイブリッドが、DNA 複製開始のシグナルとして機能する可能性を検証し、そのメカニズムと生物学的意義について解明する。具体的には、大腸菌のG4/RNA-DNA ハイブリッド依存的複製システムを用いてG4 形成が実際に複製開始に必須かを検証し、複製開始に関与するタンパク質-G4 相互作用を解明する。更にこの複製システムの、生物学的・進化的意義の解明を目指した実験を行う。又、真核細胞においてもG4 依存的DNA複製の可能性を検証する。更に、これらの研究から同定した要素を組み合わせて複製開始を担うことのできる人工レプリコンを開発する。

Outline of Annual Research Achievements

典型的な非B型DNA構造である、グアニン4重鎖(G4)構造は、生物種を問わず普遍的に存在し、その生物学的機能も次々と報告されている。G4は、配列ではなく、形成する核酸形態が重要な役割を果たす。又、G4の形成と崩壊は、生体内でダイナミックに変動している可能性があり、これまで見逃されている重要な非ゲノム情報である。数年前にヒトを含む高等生物の複製起点の近傍に高い頻度でG4形成配列が存在する事が明らかとなり、G4の複製開始における役割が示唆された。私たちは、G4がDNA複製開始のシグナルとして、あるいは、複製を抑制するクロマチン形成の拠点として機能することを発見した。本研究では、非ゲノム情報としてのG4の細胞内でのダイナミックな形成が、DNA複製開始をどのように制御するか、またその存在がゲノムの安定な維持継承にどのような影響を及ぼすかを解析し、その分子機構と生物学的意義の解明を目指す。2023年度、転写プロモーターを染色体上のG-rich配列(G4形成配列)の上流に組み込むことにより、染色体全体の複製を維持できる可能性を見出した。また、その複製開始のメカニズムを、in vitroのDNA複製系を用いて解析した。さらに、この複製系は変異を誘導しやすく、それにより環境への適応度を増加させている可能性を示した。今後、同様な複製システムが真核細胞で機能しているかどうか否か、また、生物におけるRNA-DNA ハイブリッド依存的のDNA複製システムの生物学的意義をさらに解析を行い追究していく。
