Research Abstract

メリステム発達は、多くのシグナルネットワークによって制御されており、植物ホルモンと細胞骨格成分が関わっている。しかし、メリステム発達過程におけるこの二つの要素の相互作用については、今のところ不明瞭である。本研究は、メリステム発達過程における細胞骨格成分であるアクチンと植物ホルモン間でのクロストークの解明を目的とした。我々は、a)アクチンはオーキシン応答を調節することで根のメリステム発達を調節している、b)局所オーキシン勾配はメリステム発達過程でのエチレンおよびサイトカイニン応答を調節している、という2つの仮説を立て検証した。これらの仮説を検証するために、栄養成長段階に属するシロイヌナズナ・アクチンアイソバリアントの単一および二重変異体を用いて遺伝学的解析を行った。アクチンアイソバリアントであるACT7とACT8変異体の根の成長および伸長解析から算出した細胞生産率の結果から、ACT8ではなくACT7が主根のメリステム形成を強く制御していることが判明した。また、メリステム発達に対する限定要因であるオーキシン応答の遺伝子および細胞レベルの解析から、主根の根端メリステムでは正常なオーキシン最大量を維持するのにACT7が必要であるという結果を得た。オーキシン輸送体の細胞膜局在および膜輸送系の解析から、ACT7は根端メリステムで正常なオーキシン勾配形成に必要であるPIN1とPIN2の活性を特異的に制御していることを証明した。さらに、分子生物学的および生理学的解析により、メリステムにおける異なったオーキシン勾配がサイトカイニン応答とエチレン応答に影響を与えていることが判明した。これらの結果は、植物ホルモンのクロストークに制御されているオーキシン勾配を調節することによって、アクチンアイソバリアントであるACT7が主根のメリステム発達を制御することを示唆している。
