Research Abstract

多細胞生物である高等植物の形態は、個々の細胞を取り巻く堅い細胞壁の存在により、これに規定された各細胞の分裂と生長の方向に基づいて決定されている。植物メリステムにおける細胞分裂は細胞形態にとって始原的意義をもつが、植物細胞では動物細胞とは異なる独自の分裂様式が知られている。また、細胞分化についても植物特有の細胞内構造に基づく分化様式が示されている。そこで、本研究ではメリステムにおける植物細胞形態の制御機構を解明すべく、細胞分裂と分化という基本的現象における細胞内構造とその動態についての、顕微鏡画像に基づいた定量的かつ網羅的なイメージング解析を行った。特に、細胞の形態形成に関係の深い細胞核・染色体、細胞板、液胞の動態および細胞骨格系の構造的関係を重点的に解析し、メリステムでの細胞分裂から茎根の分化形態形成に至る植物細胞に特有の細胞分裂・分化様式を多角的に明らかにすることを目指した。
本年度は昨年度に確立した撮像システムにより、下記1)～3)について、画像解析およびシミュレーション解析に取り組んだ。1)細胞分裂期における細胞膜および液胞膜近傍のアクチン繊維量の増減を定量的に示した。また、分裂中期～後期の表層アクチン繊維の動きを定量解析した。2)表層微小管の挙動を擬似乱数によって模擬するシミュレーションソフトウェアを作成し、表層微小管の配向が細胞形状や微小管形成中心の影響を受ける可能性について検討した。3)細胞外への輸送経路が葉の表皮細胞の湾曲部形成に関与する可能性を検討すべく、ゴルジ体シス層マーカーであるERD2-GFPを用いてゴルジ体の動態を定量評価した。
