可視化とオルガネラ機能解析から探る植物の機能発現メカニズム

Research Abstract

タバコにおけるエンドサイトーシス,の可視化と,そこで機能する分子の特定および機能解析を行うべく研究を継続した.その結果,エンドサイトーシスが誘導された際にエンドソームとトランスゴルジネットワークの中間的性質を有するオルガネラが出現することを見いだした.また,この過程に複数のRAB11ホモログが関与していることも明らかにした.陸上植物でRAB11ホモログは極端に多様化しているが,我々の研究結果により,この多様化が植物のトランスゴルジネットワークやエンドソームの機能発現に深く関わっていることが示された.
ゼニゴケを用いた解析においては,ゼニゴケのRAB5ホモログの機能解析に進展があった.ゼニゴケにも被子植物と同様に2種類のRAB5が存在する.このうち動物のRAB5のオルソログであるMpRAB5は液胞輸送経路において機能することが示された.この結果は,保存形RAB5の機能が陸上植物で良く保存されていることを示している.一方,植物特異的なRAB5であるARAグループについては,陸上植物においてその機能が大きく多様化していることが示された.シロイヌナズナのARA6がエンドソームから細胞膜への輸送を制御しているのに対し,ゼニゴケのARA6ホモログ(MpARA6)は葉緑体への輸送に関与していることが明らかとなったのである.これらの結果から,新たなRAB GTPaseの獲得が,植物における膜交通経路の多様化に深く関与していることが改めて示された.
