Research Abstract

ゲノムインプリンティングは、胎盤を保有する哺乳類に特異的な現象で、胎盤形成に存在の意義があると推察されている。本研究では、生殖細胞系列のエピゲノム基盤として、マウスインプリント領域(DMR)の系統的単離・同定と領域内分子機構の解明、ヒトへの保存性を基に、ヒト生殖補助医療(ART)がゲノムインプリンティング機構に与える影響と異常を起こす分子機構について明らかにすることを目的とする。さらに、胎盤特異的にインプリンティングを受ける遺伝子についても、研究を発展させる。本年度の成果は、(1)メチル化インプリント領域(DMR)における男性不妊症時患者精子のメチル化異常:ヒト男性精子のインプリント異常について、網羅的な解析を正確に行い、その異常の頻度、程度、影響を受けやすい領域を明らかにした。その結果、およそ25%にメチル化異常を認め、精子数の少ない乏精子症精子では、その頻度が高い事が判明した(80%)。同様に、程度も精子数と負の相関を示した。また、異常のパターン分類により、DNAメチル化異常は精子成熟過程の異常かあるいは初期化の異常であることが示唆された。(2)胎盤特異的インプリント遺伝子の単離:ART妊娠では、胎盤異常の頻度が高い。そのため、周産期合併症の頻度も高い。今回、マウスを用い、胎盤特異的インプリント遺伝子を複数同定した。また、ヒト胎盤へのインプリントの保存性も確認した。初期化異常を示すクローンマウスの胎盤は、肥大化する。我々が発見した胎盤特異的インプリント遺伝子は、このマウスの胎盤でインプリント異常を示した。
