初期発生における能動的DNA脱メチル化の制御機構の解明

Research Abstract

最近、我々を含む複数のグループにより、Tet3による5-メチルシトシン(5-methylcytosine;5mC)から5-ヒドロキシメチルシトシン(5-hydroxylmethylcytosine;5hmC)への変換が能動的DNA脱メチル化に関与することが明らかにされた。しかし、能動的DNA脱メチル化には、5hmCを介さない経路が存在することが示唆されている。そこで、本研究では、Tet3依存の能動的DNA脱メチル化経路に加えて、Tet3に依存しない能動的DNA脱メチル化経路について、DNA脱アミノ化の関与を中心に検討した。
5hmC非依存性能動的DNA脱メチル化としては、AIDによるDNA脱アミノ化の関与が考えられる。そこで、まずAIDタンパク質の受精卵における細胞内局在を免疫染色により調べた。その結果、AIDは受精卵において大部分が細胞質に、一部が核に局在することが明らかとなった。次に、AID KO受精卵において5mCおよび5hmC状態を免疫染色法により検討した。その結果、AIDKOのメスから得られた受精卵においても野生型と同様に雌性ゲノムに5mC、雄性ゲノムに5hmCが認められることが明らかとなった。このことから、AIDは少なくとも受精後に生じるゲノムワイドな能動的DNA脱メチル化に関与しないことが示された。
