Outline of Annual Research Achievements

本研究の二年目にあたる平成24年度は、これまで継続して開発してきたロジウムおよびルテニウム触媒系を駆使して、多様な基質とアルキンのカップリング反応あるいは分子内アリール-アリールカップリング反応を開発することに成功した。没食子酸やシリンガ酸、あるいはバニリン酸等、複数の水酸基やメトキシ基を有する安息香酸類は、植物中に広く含まれ、バイオマスから容易に入手できるため、将来的に有力なビルディングブロックとなりうる。そこでまず、そのようなカルボン酸の誘導体化について検討を行ったところ、ロジウム触媒および銅塩を酸化剤として用いる条件で、アルキンとの酸化的カップリングが、カルボキシル基のオルト位での炭素―水素結合切断を伴って効率よく進行し、対応するイソクマリン誘導体が合成できることを見出した。。一方、安息香酸と同様に入手容易なベンズアミド類中のアミド基も、よい配向基となることがわかっている。実際、II価ルテニウム錯体に銀塩を添加して生じるカチオン性ルテニウム触媒および銅塩存在下、ベンズアミドとアルケンとの酸化的カップリングが円滑に行えることを昨年報告した。今回、同様の条件でアルキンとのカップリング反応について検討を行った結果、オルト位の炭素―水素結合間へのアルキンの挿入が起こり、対応する三置換アルケンが選択的に生成することを見出した。さらに、安息香酸の代わりにフェニル酢酸類を用いて、不飽和化合物との酸化的カップリングの検討を行う中で、適当なロジウム触媒/銅塩酸化剤存在下、分子内脱水素アリール―アリールカップリングが脱炭酸を伴って進行し、対応するフルオレン誘導体が合成できることを見出した。
