Outline of Annual Research Achievements

本研究では基底状態相互作用ならびに励起状態挙動に関する詳細な情報が入手可能なBSAおよびHSAをスカッフォールドとし、ポリエチレングリコ-ル(PEG)による表面修飾による会合に起因する水溶性、さらに適用可能基質の制限など問題点の改善へ試み、さらにPEGの特性を活用した親疎水のバランスに優れた新規キラル・ソフトインターフェース反応場の構築も指向し、PEG修飾ウシならびにヒト血清アルブミンの超分子不斉光反応の不斉反応場としての活用について検討した。
PEG化は、BSAおよびHSAのリジン残基のアミノ基を活用し、グルタルアルデヒドにより片末端に反応性を付与した2鎖型PEG化試薬(MW=2,500x2)の2種の試薬を用いた修飾法により検討した。血清アルブミン(SA)とPEG化試薬との種々の混合比により得られたSAのPEG化率ならび得られた各種PEG化SAの高次構造およびACとの基底状態相互作用を、UV・CD・蛍光・蛍光寿命測定などにより検討した。AC存在下におけるPEG化を検討したところ、ACに対する結合能を維持したままのPEG化が可能である事を見出した。CDスペクトルを用いたPEG化BSA、PEG化HSAとACの相互作用を検討した結果、超分子不斉光反応に用いるにはPEG率10%程度が適切であると共に、反応には関与しない結合サイトはマスク可能である事が明らかとなった。加えて、PEG化によりSAの顕著な水溶性の向上が確認された。
さらに、HSAではHT/HHが11にまで向上し、高い生成物選択性を達成すると共に、82% eeで光二量体2が得られ、PEG化SAが有効なキラル反応場として機能する事を明らかとした。
以上より、PEG修飾による新規キラル・ソフトインターフェース反応場の創製に関する検討し、重要な知見と設計方針を見出し、その端緒を拓くことに成功した。
