Outline of Annual Research Achievements

本研究では、細胞サイズの液滴や小胞を取り上げ、マイクロメータ・スケールでのソフト界面の物理化学的特質を明らかにするとともに、微小空間でのタンパク質やDNAの機能制御手法を発展させることを目的としてきた。リン脂質を含む油層中に細胞サイズ液滴を形成させ、それをバルクの水層に移行させることにより、細胞サイズのリン脂質小胞を得るという実験手法が、有用であることが、本研究を通してますます明らかになってきた。
理論的解析の結果から、液滴のサイズが大きいほど油相から界面への移行は容易であるが、界面にはピン止めされる傾向をしめすことが予想された。逆に、小さい液滴ほど、油相から界面への移行は起こり難いが、界面へピン止めされることなく水相に移行するようになることがわかる。これは、実際の実験で観測されている事実と対応している。また、重要な結果として、液滴のサイズに関わらず、液滴はその大半が水相に移行した時点で、その上部が界面にピン止めされる傾向を示すことが、この理論モデルからわかる。また、単分子膜部分と二分子膜部分の相対的な安定性が移行の速度過程に決定的な影響を与えることが予想できる。例えば、荷電脂質を用いることで、二重膜小胞の生成効率が増大することも理論的に予測できる。
以上のように、界面透過法は、今後多様な生体高分子や基質を生体濃度で封入して、細胞サイズのリン脂質膜小胞を構築する汎用的な方法論へと発展してゆくものと期待される。
