Outline of Annual Research Achievements

生体膜の融合、分裂は生命活動の多くの過程（細胞でのベシクルによる膜動輸送、シナプスでの神経伝達物質の放出、ウイルスの感染など）で起こる素過程であり、その融合経路を明らかにすることは生物物理学として非常に重要である。本研究では分子シミュレーションを用いて、膜融合を起こす条件とその融合経路とダイナミクスを研究する。
本年度は、本研究者がこれまでに開発した粗視化脂質分子模型を2成分系に拡張し、ミセル形成、界面活性剤吸着による脂質ベシクルの分裂について研究した。まず、コーン型の洗剤型の界面活性剤と円筒型の脂質を混ぜるとバイセルと呼ばれる円盤状のミセルが形成されることを示した。コーン型と円筒型の脂質の混合系では条件によって、ヒモ状ミセルと円盤状ミセルが結合した蛸状ミセルが形成されることを明らかにした。このようなミセルはブロックコポリマー系では観察されていたが、この研究によって脂質でも形成可能であることが示唆される。
脂質ベシクルを洗剤の溶液内に入れ、洗剤型の界面活性剤分子がベシクルに吸着することによるベシクルの形態変化を調べた。界面活性剤分子の吸着によって、２分子膜に自発曲率が生じ、ベシクルがひょうたん状に変形した後、２つのベシクルに分裂、もしくはヒモ状ミセルに形態変化することを示した。洗剤分子の親水基の相対的なサイズを大きくするとヒモ状ミセルが形成する確率が増加する。
