高エネルギー構造の解明による蛋白質構造揺らぎの可視化

Outline of Annual Research Achievements

蛋白質構造において、天然状態を逸脱した高エネルギー状態の存在が溶液NMR法で確認されているが、構造やダイナミクス、安定性の解析、機能的意義の解明は天然状態ほど進んでいない。蛋白質の高エネルギー状態を圧力下で安定化させ、NMRによる立体構造を行い、最安定状態から変性状態までの一連の構造変化を可視化する事ができる。これまでに高圧力NMR法によりユビキチンには、最安定状態N1の他に準安定状態N2や局所変性状態I、完全変性状態Dが存在することを明らかにしてきた。また3000気圧下でN2構造を78%まで分布させ、立体構造解析に成功している。N2構造の精密構造解析や、機能的意義の研究のために1気圧下でN2構造が安定化された変異体を合理的に設計することを試みた。
1気圧下でのN1構造と3000気圧下でのN2構造の立体構造比較から、I36-Q41間の水素結合長とK11-E34間の塩橋が大きく変化していることが明らかになった。そこで、水素結合と塩橋の相互作用を減弱させるためにQ41A, Q41N, K11A, E34A変異体を作成し、高圧力NMR測定を含む各種NMR測定に基づいて、立体構造、ダイナミクス、熱力学的安定性の解析を行った。
圧力(3000-30 bar)による化学シフト変化と変異による化学シフト変化に相関がみられたことから、加圧時と同様に変異によってもN2構造の安定化が示唆された。最も化学シフト変化量が大きいQ41Nについて、高圧力NMR法によりN2構造の分布率が70%程度と分かった。距離情報(NOE)と角度情報(RDC, TALOS+)からQ41Nの精密な立体構造に成功した。Q41Nは、8-44-70からなる疎水性パッチの構造は保持しつつも、高圧下で決定されたWTのN2構造と同様に、α-helix, loop, β3-strand, β5-strandの配向が変化していた。
