Outline of Annual Research Achievements

孤発性筋萎縮性側索硬化症(ALS)脊髄運動ニューロンでは,AMPA受容体のサブユニットGluA2に本来行われるべきRNA編集が不充分で、この分子変化はAMPA受容体のCa2+透過性亢進を通じて運動ニューロン死の直接原因であることから、孤発性ALSの病因と密接に関連すると考えられる。GluA2のRNA 編集は、adenosine deaminase acting on RNA 2 (ADAR2) により特異的に触媒されることから,孤発性ALS運動ニューロンにおけるADAR2活性低下のメカニズムを明らかにすることは病因解明、治療の分子ターゲットの同定のために極めて重要な課題であると考えられる。昨年度、30例以上の孤発性ALS患者剖検組織を用いた単一運動ニューロンの検討から、GluA2 Q/R 部位のRNA編集率低下は疾患特異的分子異常であり、選択的なADAR2の発現低下に依り引き起こされることが明らかになった。
本年度は、その下流および上流に生じている分子メカニズムの解析を進めた。下流のメカニズムとしては、TDP-43の局在異常がおこること、それがCa2+透過性AMPA受容体の発現増加によるCa2+依存性プロテアーゼであるカルパインの活性化、カルパインによる特異的TDP-43切断、切断による易凝集性断片の形成、それがコアとなっての封入体形成、というカスケードがin vitro実験系を用いて明らかにした。さらに、患者の病的組織にもカルパインの活性上昇及びカルパイン依存性TDP-43断片が見出されたことから、ALS運動ニューロンにも同様のカスケードが働いていることが想定された。上流のメカニズムとしてADAR2 の発現低下が遺伝子の転写活性低下であると考えられるため、ADAR2 遺伝子プロモーター領域の、ルシフェラーゼを用いたレポーターアッセイ系を確立した。
