Outline of Annual Research Achievements

特定の細胞群が原因不明の変性をおこして脱落し、冒された脳の部位の違いによって様々な症状を呈する神経変性疾患は、症状が「進行性」に悪化することが重要な特徴である。神経病理学、生化学解析から、変性する部位の神経細胞やグリア細胞に疾患に特徴的な異常タンパク質を構成成分とする病変が認められ、その病変分布や広がりが患者の臨床症状と密接に関係していることが示されている。しかしながら、病変の細胞特異性や選択性、経過とともに悪化する「進行性」についてはほとんど議論されてこなかった。
申請者は、細胞内で生じた異常タンパク質が、その特殊な構造から、シナプスを介して細胞間を移動し、自身を鋳型に自己複製しながら、癌細胞やウイルスのように細胞から細胞へと伝播して広がることにより、系統的、回路選択的な神経変性がおこり、それが徐々に進行するという仮説を提唱している。
今年度は野生型マウス脳内に線維化したリコンビナントαシヌクレイン、または可溶性αシヌクレインを注入し、αシヌクレイン蓄積がみられるか検討を行った。αシヌクレイン線維接種群では接種後３ヶ月でリン酸化αシヌクレイン陽性の病理構造物が出現し、接種後１５ヶ月では接種したほとんどすべての個体でリン酸化αシヌクレイン陽性のレビー小体様構造物が認められた。この病理構造物は抗ユビキチン抗体、抗p62抗体陽性であった。主な蓄積部位は海馬歯状回、CA1-3、扁桃体、線条体、感覚野などであり、接種部位から離れた部位にも蓄積が認められた。一方、可溶性αシヌクレイン接種群ではレビー小体様構造物の出現は認められなかった。以上の結果は、αシヌクレイン線維の接種により、野生型マウスの脳内の内在性αシヌクレインが異常に変換され、それが広がることを直接証明する結果といえる。
