母体の食変化と子の脳機能発達に関する病態神経科学研究

Outline of Annual Research Achievements

これまでに、母体の妊娠前～離乳までの高脂肪食摂取は、幼若期産仔海馬で酸化脂質の蓄積と神経新生低下、BDNF値低下、樹状突起分岐不全、記憶学習の低下を誘導することをマウスで見いだしている。そこで、本新学術領域の研究では、当該の影響がシナプス後部構造であるスパインを中心とした脳の微細形態とその動態にどのような影響を及ぼすかを、マウス個体レベルで明らかにすることにした。具体的に、離乳後も産仔で高脂肪食摂取を継続した場合、または離乳後は通常食を摂取した場合、それぞれにおけるスパインへの影響を検討した。用いた研究手法はin vivo 2光子イメージング等による機能形態学的解析である。マウスはThy1-YFPマウスを使用した。その結果、　離乳後も高脂肪食摂取を継続した産仔マウスのみならず、離乳後通常食を摂取した産仔マウスにおいても、週齢を重ねるごとに、海馬および内側前頭前皮質において、樹状突起スパインの数が減少し、またスパインの成熟遅延が観察された。これまでの迷路学習試験では成長後記憶学習能の変化は消失することが判明しているが、今回の結果は、母体の高脂肪食摂取による影響が、病状（表現型）の検出には至らないまでも、スパインにおいて継続し、潜在的な神経機能異常が残存していることを示唆する。かつ、スパインの変化が海馬以外の領域においても検出されたことは、母体の影響は産仔において広範囲の脳領域に及んでいると考えられる。今回観察された非顕在性のシナプス機能形態学的変化が成長後の疾患罹患性に影響するか、今後検討を行う。また、培養細胞においても、酸化脂質処理により初代培養神経細胞のスパインが減少することが確認されたので、今後は、離乳後の食環境の変動が脳機能発達においてどのような形で反映されるか、その詳細を複数のアプローチで明らかにする予定である。
