Outline of Annual Research Achievements

神経細胞は1本の軸索と複数の樹状突起を形成して極性を獲得する。「対称性の破れ」は神経極性形成の最初のステップである。我々はこれまでに、定量的なライブイメージングと数理モデルを組み合わせて、培養神経細胞がShootin1により自発的対称性の破れを引き起こす分子機構を明らかにした。神経細胞は、非対称な細胞外シグナルが存在しない培養条件下ではランダムな方向に軸索を形成するが、脳組織内においては特定方向に整然と軸索を伸ばす。本研究は、Shootin1が引き起こす神経細胞の自発的対称性の破れに方向性をという拘束を与える細胞外シグナルを同定し、内在性の対称性の破れの機構と非対称な細胞外シグナルとのクロストークを明らかにする。さらに、細胞内Shootin1のゆらぎが、神経細胞の対称性の破れと極性形成に積極的に活用される可能性を検証する。
昨年度は、神経細胞の自発的対称性の破れに方向性を与える細胞外シグナルNetrin-1がPAK1によるShootin1のリン酸化を介してクラッチ効率を高め、軸索形成促進を引き起こすことを見出した。今年度は、細胞内Shootin1のゆらぎが、神経細胞の対称性の破れと極性形成に積極的に活用される可能性を検証した。Shootin1の神経突起先端における濃縮にはノイズを伴う大きなゆらぎが観察された。そこで、定量的な数理モデルを用いてこのゆらぎを徐々に規則的にしてゆくと、極性化に失敗して、本来1本のみ形成すべき軸索を複数の形成する神経細胞の割合が急激に増加することがわかった。以上の結果により、Shootin1のゆらぎが、神経細胞の対称性の破れと極性形成に積極的に活用されて「強固性」や「正確性」をもたらす可能性が示唆された。
