Outline of Annual Research Achievements

A：マウスジェネティクスを駆使した異常骨髄ニッチモデルの解析
1.骨前駆細胞特異的AML1欠損マウス（Col1-Cre-AML1 fl/flモデル）の樹立および2.骨前駆細胞特異的AML1欠損マウスのフェノタイプ解析において、①造血幹細胞プールサイズ、自己複製能、細胞周期、細胞死の解析　②骨前駆細胞のフェノタイプの解析及び骨前駆細胞発現AML1の下流遺伝子単離　③骨前駆細胞特異的AML1欠損マウスの骨髄異形成症候群や白血病移行の検証、異常造血細胞の由来検索、異常造血細胞移植実験（造血系再構築、二次性白血病発症の検証）を行ったところ、RUNX1cKO及びInv16cKIのマウスジェネティクスで、RUNX1欠失にて大幅に白血病発症率の低下を認めること、一方骨髄微小環境においてはRUNX1欠失にて骨髄幹細胞（KSL分画）が著明に減少するが、それだけでは白血病発症は認めない事を見出した。B：骨前駆細胞培養実験系の構築と骨髄ニッチの可視化については、1. 新規骨前駆細胞・造血幹細胞共培養システムを用いた実験　2. レトロウイルスshRNAiにより改変された骨前駆細胞・造血幹細胞共培養実験を行い完成した。
B：ASXL1コンディショナルミューテーションノックインマウスの樹立
現在ASXL1コンディショナルノックインターゲットES細胞を樹立し、既にキメラマウスを樹立完成したことから目標はほぼ達成した。。ミューテーションはヒト白血病症例に高頻度に認めれるEXON12に設定した。今後ヘテロマウス、ホモマウスを樹立し、ASXL1の正常造血における働き、ミューテーションによる白血病発症を再現し、さらなる白血病発症機構の解明を行う。
