Outline of Annual Research Achievements

ITCは主に骨髄の場において生物学的な活性が段階的に修飾され、転移しない良性ITCから転移能を獲得した悪性ITCに変化することが明らかとなった。その過程においては宿主ニッチ・微小環境（筋線維芽細胞・骨芽細胞・食細胞）からのサイトカイン等の刺激によりITCの悪性化に際してエピジェネティックな修飾が蓄積され、５－６個の内因性マイクロRNA発現に特徴ある変化が現れる。さらに細胞治療学的な手法により、特殊な配列を有する人工合成マイクロRNAを細胞に導入することにより細胞形質を大きく変換できる技術を整備した。【骨子】消化器癌の転移を左右する骨髄内遊離癌細胞のマイクロRNAを解明し、癌転移の早期診断および介入創薬に向けて、未来型医療の基盤を構築した。今年度は特に、臨床像と有意の相関あるマイクロRNAに絞りこみ、その病態的意義を解明した。【結果】本年度は当初の計画に基づいて、消化器癌患者の骨髄細胞につき、倫理委員会等の必要手続きを申請および承認を得た後に採取を実施し、高感度細胞分離装置で解析した。その結果、転移を有する患者とない患者のCD45(-)EpiCAM(+)の骨髄内上皮細胞を比較して特徴的なマイクロRNA分子が発現していることを明らかにした。今年度の解析の結果、固形癌患者で転移系性能を左右するマイクロRNAの一群は嫌気性解糖系制御に関わる情報伝達経路に集中的にマップされた。【考察】消化器癌患者の骨髄では、発現マイクロRNA分子種が上皮系パターンに偏倚していた。興味深い事に、造血系の細胞でも上皮系マイクロRNAを介した細胞の上皮化現象が行われており、この現象が原発巣の消化器癌からのケモカインおよび血中マイクロRNAによる遠隔効果である可能性が推測された。今年度の研究成果で転移を左右する癌微小環境の特徴をマイクロRNAの視点から究明し、創薬シーズを提示した。
