Research Abstract

私たちは、上皮基底膜の重要な接着分子ラミニン332やその構成鎖の一つであるγ2鎖(Lm-γ2)が、がんの悪性進展を支える重要な微小環境因子と考え、その作用を調べてきた。Lm-γ2は、がんの浸潤先進部で単独で過剰に発現し、またその強制発現によって浸潤性増殖が促進される。本研究では主としてこのLm-γ2の作用機構の解析を進めた。その結果、Lm-γ2やそのN末端断片が血管内皮細胞の収縮を促進することによって、血管透過性を高めたり、がん細胞の血管内皮細胞層への侵入を促進することを見出だした。これらの結果は、浸潤性がん細胞が過剰に産生するLm-γ2が腫瘍血管に作用し、がんの転移や増殖を促進する可能性を示唆する。さらに、Lm-γ2の受容体を探索した結果、Lm-γ2が、がん幹細胞の重要なマーカー分子であるCD44と直接的に相互作用すること、またCD44の細胞内ドメインのリン酸化を促進し、CD44依存的にがん細胞の移動を促進することを見いだした。この結果は、がん浸潤マーカーであるLm-γ2が、がん幹細胞マーカーCD44と協調的にがんの悪性化やがん幹細胞機能の維持に関与する可能性を示唆する。
一方、私たちは過去に腫瘍血管に特異的に発現する接着タンパク質angiomodulin (AGM)/IGFBP-rP1を見出だしており、本研究ではその発現調節機構や血管内皮細胞に対する作用を調べた。その結果、AGMが内皮細胞においてVEGFにより強く誘導されること、AGMがインテグリンαvβ3に作用することによって内皮細胞の接着を促進することを見いだした。これらの結果からAGMが腫瘍血管の異常な構造や機能に関係することが示唆される。
