Outline of Annual Research Achievements

スキルス胃癌細胞44As3の播種性浸潤における腫瘍細胞と間質線維芽細胞の相互作用について解析を行った。44As3細胞と間質線維芽細胞を３次元マトリゲル上で共培養すると、両細胞からなる特徴的な浸潤性の細胞凝集塊が形成された。この細胞凝集塊においてSFは浸潤先端を含む中心部に、44As3細胞はその周りに存在していた。この細胞外基質リモデリングの機構をより詳細に解析するため、蛍光ゼラチンでコートしたカバーガラス上で両細胞を共培養した。その結果、間質線維芽細胞はゼラチンマトリックスの破壊を軽度に起こすが、44As3細胞との共培養によりこの活性が顕著に促進されることが明らかになった。一方、44As3細胞のみの培養ではゼラチンマトリックスに変化は認められなかった。また、間質線維芽細胞の収縮能を阻害するミオシン阻害剤で処理すると、両細胞によるゼラチンの破壊や３次元マトリゲルの浸潤が抑制された。同様にSrcキナーゼの阻害剤もこのような細胞外基質のリモデリングを阻害した。以上の結果から、スキルス胃癌細胞は、線維芽細胞との相互作用により細胞外基質の物理的なリモデリングを行い、これが浸潤性に寄与することが示唆された。これをイメージングにより定量化し、マルチウェルプレートを用いてがん特支援班から供与された阻害剤ライブラリのスクリーニングを行った。Src経路、Rho経路などいくつかのシグナル経路の阻害剤が異なった形で胃がん細胞とSFとの相互作用に影響をあたえることがわかった。Src阻害剤についてはマウスの腹膜播種モデルにおいて腹膜播種巣の形成を阻害し、形成された播種巣の組織像では間質線維芽細胞の腫瘍組織への侵入が阻害されている像が観察された。
