Outline of Annual Research Achievements

驚くほど多数の老人性難聴の患者が存在するにもかかわらず、内耳の機能異常・病態の研究は、内耳が微細な構造体であることから、十分に進んでいない。本研究では、内耳特異的KOマウスを用いて、イメージング手法(個体~1分子)を駆使することにより、内耳疾患の新規治療法の開発に繋がるような新しい研究法を確立しようというものである。
本年度は進行性の高度難聴(80 dB)を呈する内耳有毛細胞特異的Cdc42ノックアウト(KO)マウスを作製し、難聴発症機序の解明を試みた。Cdc42 KO聴覚有毛細胞の電子顕微鏡解析では、聴毛の欠落・癒合が認められた。蝸牛の有毛細胞ではGFP-Cdc42は、聴毛を覆う形質膜と頂側に近い側壁に局在した。聴覚有毛細胞のモデル細胞として用いたMDCK細胞では、1) GFP-Cdc42はcyst管腔に面する頂側面と頂側に近い側壁に局在し、2) Cdc42-ノックダウン(KD)安定細胞株では、微絨毛の減少や癒合とTight junction (TJ) のマーカーであるZO1の細胞内停留が認められた。2) TJの形態異常はKOマウスの聴覚有毛細胞(電顕)でも確認できた。Cdc42 FRET biosensor TG miceの聴覚有毛細胞を用いた二光子蛍光顕微鏡観察では、Cdc42は、聴毛膜の先端部で強い、頂側に近い側壁で弱い活性化を示した。Cdc42-KD細胞株における細胞内シグナルに関しては、CofilinとMyosin phosphatase 1 (MYPT1) のリン酸化亢進に代表されるアクチン代謝障害と代償経路の活性化 (RhoA→ROCK→ LIMK2→Cofilin, RhoA→ROCK→MYPT1→MLC) が観察された。
以上より、Cdc42 KOマウスの進行性難聴は、聴覚有毛細胞内の聴毛及びTJでのアクチン代謝障害に起因することを明らかにした。
