Outline of Annual Research Achievements

本研究では、ショウジョウバエ腫瘍悪性化モデルを利用し、エンドサイトーシス制御破綻が引き起こすがん悪性化機構を遺伝学的に解析した。ショウジョウバエ複眼原基の上皮組織において、遺伝的モザイク・クローン法によりがん原性Ras (RasV12) を発現する体細胞クローンを誘導すると、これらの変異細胞クローンは過剰に増殖して腫瘍を形成する。このRasV12誘導性腫瘍は浸潤性や他器官への転移能を示さないことから、良性腫瘍と見なすことができる。われわれは、このRas誘導性の良性腫瘍に対して一連のエンドサイトーシス制御遺伝子の突然変異を導入すると、変異細胞が浸潤・転移能を獲得することを見いだした。この浸潤・転移能はエンドサイトーシス制御遺伝子の変異のみでは付与されないことから、Rasシグナルの活性化とエンドサイトーシス制御破綻の協調により上皮の腫瘍悪性化が引き起こされたと考えられた。興味深いことに、エンドサイトーシス制御遺伝子rab5の機能を欠損したRas活性化細胞（RasV12/rab5-/- 細胞）は浸潤・転移能を獲得するだけでなく、その周辺細胞の増殖を促進することが分かった。そこで、RasV12/rab5-/- 細胞が浸潤・転移能を獲得する機構、および周辺細胞の増殖を促す機構の遺伝学的解析を進めた結果、RasV12/rab5-/- 細胞内においてJNK経路を介してがん抑制経路Hippo経路が抑制され、これによりUnpaired （Upd; IL-6ホモログ）の発現誘導が引き起こされることが分かった。さらに、Upd によるJAK/STATシグナルの活性化が、RasV12/rab5-/- 細胞自身の浸潤・転移能獲得と周辺細胞の増殖促進の両方に必要であることが分かった。
