Outline of Annual Research Achievements |
[1] 大腸癌臨床検体では浸潤部においてヒストンH3の9番目のリジンのトリメチル化 (H3K9me3) が有意に亢進しており、H3K9me3の量がリンパ管侵襲と相関する事を見出した。さらにin vitroではH3K9メチル化酵素SUV39H1の活性は、細胞遊走能を制御し、モデルマウスを用いた実験ではSUV39H1の過剰発現が腫瘍形成を促進し、生存率を低下させる事を明らかにした。乳癌において発現が亢進するEGF様増殖因子amphiregulinは細胞膜から核膜へ局在変化し増殖因子領域非依存的にH3K9me3を亢進し、細胞遊走を活性化する事を見出した。 [2] 本研究において私達は、SUV39H1は遺伝子発現制御を介さず、ゴルジ体構築を調節し、細胞極性形成に影響を与え、細胞運動を調節することを明らかにした。さらにヒストン修飾状態をゴルジ体へ伝える分子として、核膜貫通タンパク質SUNおよびnesprinからなるLINC複合体を見出した。またLINC 複合体に結合するゴルジ体関連タンパク質群を見出した。 [3] クロマチンシグナリングを伝達する核膜構築分子基盤の解明するため核膜内膜タンパク質 lap2beta;を利用して、核膜局在化アッセイ系を構築し、その局在化には可溶性因子とATPが必要である事を明らかにした。 本研究において、グローバルなヒストン修飾が、LINC複合体を介して、ゴルジ体構築を制御し、その結果、細胞極性形成、細胞運動活性を調節していることを明らかにしてきた。さらに、ゲノムワイドなH3K9me3状態は細胞運動を調節し、癌の進行に重要な役割を果たしていることも示した。これらの結果は、遺伝情報発現の場である細胞核が細胞内外の情報を統合して、クロマチン構造を利用して、細胞機能を調節していることを示しており、細胞の高次生命現象に細胞核が担う機能の統合的な理解に大きな意義を持つ。
