Research Abstract

ゼブラフィッシュの胚・仔魚を対象にして、形態的に相同な後脳の網様体脊髄路ニューロン群(RSNs)のなかで、M細胞だけが特異的な単発発火パターンを獲得する点に着目して、そのメカニズムを解析した。その結果、M細胞は発生初期には他のRSNsと同じように連続発火を示すが、ゼブラフィッシュが聴覚に応答し始める頃に発火特性を大きく変えることを見出した。さらにその分子基盤には2つの低閾値型カリウムチャネルの発現が重要であることを明らかにした。本年度はKv1.1サブユニットで構成される低閾値型カリウムチャネルの発達過程における発現、および補助サブユニットKvβ2の発現を解析し、それら2つのサブユニットの組み合わせがM細胞特有の単発発火に必要であることを証明した。KvβサブユニットがKv1.1チャネルの膜表出を促進するというメカニズムは今までにない知見である。研究成果はJournal of Neurophysiology (2014)に発表し、国内外で高く評価された。M細胞と他のRSNsと結合様式をゼブラフィッシュと同じRSNsを持つキンギョで調べたところ、M細胞からそれらのRSNsへ一方向の結合があり、結合様式と形態的相同性の間に密接な関係を見出した。すなわち、分節に繰り返される相同ニューロンにはM細胞から同じ結合様式が繰り返されるのである。その成果はThe Journal of Neuroscience (2014)に発表し(我々の図が雑誌の表紙を飾った)、国際的に高い評価を得た。
