Outline of Annual Research Achievements

機能的に重要な役割を担う神経回路の形態学的な特徴を明らかにすることを目的に、脳内で頻繁に活動する神経回路を可視化するための要素技術の確立を目指した。歯状回顆粒細胞―ＣＡ３錐体細胞間の回路網とＣＡ３―ＣＡ１錐体細胞間の神経連絡に焦点を当て、シナプス結合や分子発現に関する活性化回路の解剖学的特徴を明らかしながら、１）活動の高い細胞群は幾つの回路に分類できるのか？２）活動レベルの高いシナプス後神経細胞には幾つの活動レベルの高いシナプス前細胞が存在するか？等、脳内活動回路に関する基本的な疑問の答えを求め、メゾ回路の実体に迫りたいと解析を進めてきた。
本研究課題は、大別して実験動物作製とメゾ回路の解剖学的解析の２本立ての計画で、平成23年度分の動物作出実験は予想通り進み、回路の解析は遅れた。そこで24年度は神経連絡の解剖学的解析を中心に推進した。しかし、この解析の初期段階で、活動神経回路の全体を明瞭に標識し、活動レベルの高低を電子顕微鏡下で判別できる様にすることが難しいことが判明した。
そこで明瞭に標識された一部分の神経組織のみを解析対象にして、その中に含まれる神経細胞の微細構造を特徴付けし、活動神経回路が持つ機能ドメインの形態的特徴を明らかにすることにした。具体的にはシナプス前後の活動標識の有無によりシナプスを分類し、そのシナプス後肥厚の大きさ（面積）を計測した。この結果、活動マーカーがシナプス前後に陽性のシナプスは、片方が陽性のシナプスに比べて有意に大きいシナプス後肥厚を持ち、前後共に陰性のシナプスは小さいことを示す解析結果を得た。この結果は、高い活動レベルの神経細胞どうしはより機能的に成熟したシナプス構造で結合し、機能的にも強く結びついていることを示唆している。
