精子形成周期依存的な精子幹細胞制御機構の解析

Outline of Annual Research Achievements

セルトリ細胞で観察されるステージ依存的な遺伝子発現変化は、複数のシグナルがステージ依存的に活性化することに制御されていると考えられる。我々は、レチノイン酸シグナルとFGFシグナル系がGDNFの発現パターンや未分化精原細胞の増殖パターンと相関あるいは逆相関して活性化することを見出した。今年度は特にFGF シグナルの下流で機能すると考えられるMEK-ERKシグナル系に関する詳細な解析を行った。阻害剤を用いたin vivo,及びex vivoの解析及びERK1/2のノックアウトマウスの解析から、ERKシグナルはセルトリ細胞で周期的に活性化しており、多くの遺伝子制御にかかわっているが、特にGDNFを正に制御しており、その結果として未分化精原細胞の増殖を推進することがわかった。またERKシグナルは未分化精原細胞の中でも最も幹細胞活性の高い、GFRa1細胞で活性化しており、細胞の増殖に必要なGFRa1やRet遺伝子の発現を促し、一方細胞の分化を抑制する機能をもつことが明らかになった。興味深いことにやはり未分化精原細胞で細胞の分化抑制機能を持つNanos2とは異なる機構で働くことがわかった。またGS細胞を用いた解析からERKはこれまで主に細胞増殖に関与することが示されていたAKT経路とは独立の機構で機能していることも明らかになった。以上、精子周期に特異的なERK活性がセルトリ細胞で観察され、また主に分化を促進するレチノイン酸シグナルと発現及び機能の面で逆の効果をもつことから、これらのシグナル系の協調的作用により、精子周期が制御されていることが明らかになった。
