Outline of Annual Research Achievements

本研究は、ポリ(ADP-リボシル)化酵素タンキラーゼによる蛋白質の翻訳後修飾シグナルが、染色体分配および細胞運動をどのように制御し、その破綻がどのようにがん悪性化形質をもたらすかを解明することを目的とする。我々はこれまでに、タンキラーゼの過剰発現がテロメアタンパク質TRF1のポリ(ADP-リボシル)化を亢進させ、TRF1のユビキチン分解を導くことにより、がん遺伝子オーロラAの過剰発現による細胞分裂異常を抑制することを見出してきた。この成果から、オーロラA によるTRF1の異所性リン酸化が微小管による動原体の捕捉を阻害することが示唆された。そこで今年度は、微小管による動原体捕捉に対するTRF1そのものの機能的関与について詳細に検討した。まず、TRF1の枯渇は、微小管による動原体の捕捉を過度に増強した。分裂期キナーゼの一種であるオーロラBは、自身のキナーゼ活性に依存し、動原体における微小管の脱重合の促進、すなわち異常な動原体捕捉の解消に寄与しているが、免疫染色の結果、TRF1枯渇細胞では分裂期におけるオーロラBの動原体局在が顕著に減弱していることが明らかとなった。これらのことから、TRF1は微小管による動原体の捕捉を適切に調節することにより、染色体安定性の維持に寄与していることが示唆された。一方、我々が同定したタンキラーゼ結合蛋白質TAB182を枯渇させると、細胞の運動能が亢進した。TAB182がタンキラーゼによってポリ(ADP-リボシル)化を受けることを試験管内の検討で明らかにしており、今回、タンキラーゼとTAB182の相互作用およびポリ(ADP-リボシル)化反応が、細胞運動を制御している可能性を見出した。これらの分子基盤をさらに詳細に解明することにより、ポリ(ADP-リボシル)化修飾シグナルを標的とした、新たながん治療法への道筋が開拓されると期待される。
