Outline of Annual Research Achievements

我々は昨年までに酸化ストレス依存性にERK/Fra-1経路を介してIL-11の発現が誘導されることを明らかにした(Nishina et al, Sci Signal 2012)。今回我々は環境汚染物質中に含まれる親電子物質の1種である1,2-Naphthoquinone (NQ)とよばれる物質がIL-11の発現を効率良く誘導するという現象を見出した。その後の解析から活性酸素刺激と同じように持続的なMEK/ERK経路の活性化とその後のFra-1の蓄積が、1,2-NQによるIL-11の発現に必須であることが判明した。1,2-NQはNRF2経路を活性化することが既に報告されていることから、今回の研究により1,2-NQはNRF2経路と共にIL-11経路を活性化することで、酸化ストレス応答を誘導していることが明らかとなった。
またIL-11はヒトの胃がんや大腸がんで、またマウスのそれらのがんモデルにおいても高発現していることが知られている。そこでIn vivoにおいてIL-11産生細胞を可視化するために、BACクローンを用いてIL-11-GFPレポーターマウスを樹立した。現在一種のレポーターマウスのラインは、内在性のIL-11の発現とGFPの発現が非常によく相関していることを見出している。さらにIL-11の高発現している精巣で抗GFP抗体による免疫染色により、IL-11陽性細胞の同定に成功している。今後は、IL-11-GFPレポーターマウスに様々な炎症を誘導し、また胃がんや大腸がんモデルマウスと交配することで、IL-11産生細胞の同定を目指す予定である。
