Outline of Annual Research Achievements

網膜分化における、ヒストンメチル化の役割を検討するために、マウス網膜をさまざまな発生段階で単離し、分化にかかわる既知の遺伝子の領域のヒストンメチル化、アセチル化についてChIP解析を用いて、検討を加えた。さらに、網膜を表面抗原などでセルソーターを用いて単離し、単離したサブセットの細胞群におけるヒストンメチル化の状況をChIP 解析で検討した。その際、少量の細胞数で安定したChIPのデータがだせるよう系の改善をおこなった。 その結果、網膜の介在細胞の一種である双極細胞に注目の分化に重要な遺伝子の領域がヒストンH3K27のメチル化を網膜全体でうけること、しかし網膜の分化にともない、双極細胞の前駆細胞のみで、脱メチル化がおこる事をみいだした。その結果、当該の遺伝子が網膜のサブセットの細胞で発現が可能になり、双極細胞の分化にいたることを見いだした。その際、脱メチル化酵素であるJmjd3が網膜の介在神経特異的に発現している事をみいだした。さらにこの酵素を、マウス網膜でノックダウンすることにより双極細胞の分化が抑制されることをみいだした。一方、メチル化酵素の網膜特異的ノックアウトは、増殖の阻害が見られるが分化には大きな影響はなかった。増殖についてはArf/Ink4aの発現が著しくあがることが関与している事が明らかになった。以上のように、網膜において、細胞系列特異的な脱メチル化の重要性が明らかになった。
