Outline of Annual Research Achievements

胸腺髄質上皮細胞(mTEC)の分化プロセスにおけるAireのはたらきを明らかにする目的で、Aireのプロモーター下にCre recombinaseとエストロゲン受容体リガンド結合領域変異体との融合タンパク質(CreER)を挿入したノックインマウス(Aire/CreER-KI)を樹立した。次いで、GFPあるいはRFPレポーターマウスと交配後の成体に、4-水酸化タモキシフェン(tamoxifen: TMX)を投与することでAire発現細胞をマーキングする、いわゆる時期特異的fate-mappingを行った。それによって、以下の事実が明らかとなった。
1)mTECの成熟化に伴うAireの発現時期から、最終的にAire+ mTECが胸腺から消失するまでの期間は約3週間で、半減期は7#12316;8日であった。
2)Aire+ mTECの寿命はAireの欠損によって変化しなかった。
まず、1)の知見より、Aire+ mTECの寿命が、抗Aire抗体を用いて行われた従来の実験結果のおよそ倍であることが判明した。それは、Aire+ mTECがAireの発現とともに死滅するのではなく、Aire発現消失後の、より成熟した時期(post-Aire stage)が存在するという、私どものモデルを支持する結果である。また、Aireが成熟mTECにおいてTRA遺伝子を正に制御するとともに、apoptosisを誘導することでcross presentationを促進するというモデルがあるが、2)の結果は、そうしたAireのproapoptotic活性を否定するものであった。異常の知見は、AireがmTECの分化プロセスに重要な役割を担うことを示唆している。
