Outline of Annual Research Achievements

ATP結合タンパク質のATP結合前後における水和水の配向変化をX線および中性子回折法により原子レベルで観察し、ATPエネルギー変換を“負エントロピー”を産み出す構造変化の切り口から解明することを目的とする。ATP結合タンパク質として、ATP類似物質結合により10数個の水和水構造変化を起こすタンパク質DAPK(Death-associated protein kinase)を取り上げた。DAPKは合成DNAを用いて大腸菌で発現させ、イオン交換樹脂・ゲル濾過カラムクロマトグラフィー精製を行い、75mgの試料を取得した。結晶化剤PEG1000,硫酸アンモニウム、硫酸リチウムで結晶析出することが判明し、DAPK結晶成長相図を作成し、それをもとに結晶育成を行い、最大0.032mm3の結晶を得ることが出来た。それをX線結晶解析実験に用い、DAPK単独およびATP類似物質を結合させた状態でのリガンド結合部位のフーリエ図を解析し、リガンド結合部位付近の水分子数がそれぞれ23個,24個と求められた。中性子回折実験用大型単結晶(1 mm3以上の結晶は現時点では得られず、中性子回折実験は未だ出来ていない。DAPKにATPアナログ分子が結合するとき発生する熱を等温滴定熱量測定装置で行い、結合定数(Kd / M:5.41×10-7)、反応の結合比(n:1)、エンタルピー変化(ΔH / kJmol-1 :-57.3±0.4)、およびエントロピー変化(ΔS0 / JK-1mol-1:-72±1)をそれぞれ取得した。
