Outline of Annual Research Achievements

本申請課題では,自然免疫系の受容体によるリガンド認識機構やシグナル伝達機構を主にX線結晶構造解析により明らかにすることを目的として,主に以下の3つのグループに焦点を当てて研究を進める。
1.核酸認識TLRs (TLR7,TLR8,TLR9)と核酸の複合体(抗ウイルス薬,ワクチン)
2.細胞内NLRs (Nod1,Nod2)とリガンドの複合体(クローン病,アレルギー)
3.TLRの輸送や機能を制御する蛋白質(RP105/MD-1,PRAT4A,B)(自己免疫疾患)
(TLR4/MD-2の構造解析について)TLR4/MD-2受容体は,グラム陰性細菌の外膜構成成分であるlipopolysaccharide (LPS)を認識し,細胞内にシグナルを伝え免疫系を活性化する。過度のLPS応答は,生命を脅かす敗血症を引き起こすが,その有効な治療薬は開発されていない。Lipid IVaはLPSの生合成前駆体であり,ヒトTLR4/MD-2に対してはアンタゴニスト、マウスTLR4/MD-2に対してはアゴニストとして作用するが詳細な機構は明らかになっていない。本年度は,マウスTLR4/MD-2/lipid IVa複合体およびマウスTLR4/MD-2/LPS複合体の構造解析を成功させた。(核酸認識TLRについて)本年度は,核酸を認識する自然免疫系受容体のTLR7,TLR8,TLR9について,結晶化に適した発現系の構築を進めるとともに有望なコンストラクトについて結晶化を進めた。ヒトTLR8について、リガンド非結合型および低分子リガンド結合型での構造解析に成功した。これに関して、引き続きRNA複合体の構造解析を目指して研究を進めている。
