Outline of Annual Research Achievements

大脳新皮質の発生過程では多くの転写因子やモルフォゲンが決められた組織・時期特異的に発現するよう正確に調節されているが、その発生時期と発現する細胞を決定する分子制御機構はあまり明らかにされていない。本研究ではAmnSINE1と呼ばれる転移因子SINEの1つ(AS021遺伝子座)が大脳新皮質においてSatb2遺伝子の発現を誘導するエンハンサーであることを示し、その制御メカニズムを明らかにすることで、SINEを介した大脳新皮質の形成機構の解明を目的とした。これまでに以下の点を明らかにした。(1) AS021遺伝子座のエンハンサー機能は、大脳新皮質の発生過程で早期に生じたSatb2発現細胞で見られ、さらに脳梁伸長ニューロンにおいてエンハンサー機能を有することを見出した。(2) AS021配列に結合する転写因子を発見した。この転写因子の欠損マウスでも脳梁形成に異常が見られることが知られていることから、AS021遺伝子座は脳梁形成に関わる発現カスケードの重要な一端を担っていると考えられる。さらにその転写因子が、複数のSINE由来配列にも結合可能であることを示した。(3) AS021遺伝子座においてSINEがエンハンサー機能を獲得するに至る過程を明らかにした。これらの結果から、転写因子結合モチーフを内部に含むSINEが哺乳類の祖先のゲノム中で転移増幅し、それが配列進化の過程でエンハンサーとなることでSatb2の発現をもたらす哺乳類特異的な発現カスケードを生じて脳梁の形成に至った可能性が考えられる。さらにこの転写因子は複数のSINE由来配列にも結合することから、哺乳類の祖先における制御領域の増大と様々な遺伝子発現カスケードのリモデリングに寄与した可能性を示唆するものである。
