Outline of Annual Research Achievements

神経細胞は高度に極性化した細胞であり、その分化の過程で通常一本の軸索と複数の樹状突起を形成する。しかし、神経極性形成の最初のステップである軸索形成がいかにして制御されているのか、その分子機構は未だ十分には理解されていない。本研究課題において我々は、神経極性制御因子の一つであるPar3が神経栄養因子などの外界シグナルの刺激によってリン酸化され、軸索形成を制御していることを明らかにした。さらに、リン酸化されたPar3はモータータンパク質であるKIF3Aとの結合能が低下し、軸索内輸送されにくくなることを示した。この結果は、Par3のリン酸化による軸索内輸送の制御が神経細胞の軸索形成に深くかかわっていることを示唆している。さらに我々は、長年未解明のままであった「生体内」での軸索形成の制御機構の一端を明らかにすることができた。大脳新皮質の興奮性神経細胞は脳室帯で生まれ、中間帯に入ると共に多数の未熟な突起を持った多極性細胞に変化する。中間帯で多極性細胞は双極性細胞へと変化し、先導突起と後方突起の二本の異なった突起を伸張する。双極性細胞は最終的に大脳皮質板へと移動し、成熟した神経細胞になる。この過程において先導突起は樹状突起に、後方突起は軸索へと成熟していくと考えられている。後方突起(軸索)形成にはLKB1、CaMKKなどが関与していることが我々を含めた複数のグループによる研究より、近年明らかとなってきた。本研究課題において我々は細胞接着を介したシグナルが生体内で軸索形成に大きな役割を果たしていることを見出した。
本研究課題により、培養神経細胞レベルから個体レベルまでの解析を統合して行い、より包括的に神経細胞の軸索形成機構を解明することができた。この結果は脳損傷時の再生医療などに結びつくことが期待される。
