Outline of Annual Research Achievements

大脳新皮質の神経細胞は、興奮性と抑制性神経細胞の２種に大別することができ、抑制性神経細胞は20から30%を占めることが知られている。parvalbumin (PV) 発現細胞はその約半分を占め、電気生理学的にはfast-spiking 細胞に分類され、高次機能発現や神経・精神疾患との関連が示唆されている。我々は先行研究にて、PV細胞の樹状突起と細胞体を完全に可視化する遺伝子改変マウスの作出に成功した。このマウスを用いて免疫蛍光染色を行い、抑制性シナプス前マーカー・抑制性シナプス後マーカーを可視化した。共焦点顕微鏡にて３次元スタック画像を取得し、PV細胞へのシナプス入力様式を、第一次体性感覚野において定量的に解析した。なお、抑制性シナプス後マーカーにはgephyrinを使用したが、電子顕微鏡下で、抑制性シナプス後部に局在することを確認している。(I) まずはvesicular GABA transporterをシナプス前マーカーとして使用し、全抑制性入力を解析した。 (II) 次に、皮質抑制性神経細胞の約半分を占めるPV、20から30%を占めるsomatostatin (SOM)、10%弱を占めるvasoactive intestinal peptide (VIP) 発現細胞の軸索終末を可視化し、PV細胞への抑制性入力を解析した。PV、SOM共に、細胞体へのシナプス入力量は少なく、樹上突起を好む傾向が認められた。一方、VIP入力は細胞体を好み、樹状突起への入力は少なかった。以上より、PV細胞の樹状突起は抑制性入力の約60%をPV細胞から、細胞体は約62%をVIP細胞から受けることが分かった。すなわち、PV細胞は樹状突起・細胞体という２つのコンパートメントにおいて、抑制性入力様式が異なることが明らかとなった。現在は第一次運動野において、上記所見の普遍性について検討している。
