Outline of Annual Research Achievements

細胞集団の協調的振る舞いが引き起こす形態形成を理解するためには、細胞集団に働く力の定量化が必須である。平成24年度は、開発した力のベイズ推定法(Ishihara and Sugimura, 2012)について、その正確性と安定性の比較評価を行った。その結果、比較検討した3つの手法の中で、我々の力のベイズ推定法が最も正確で、かつ、画像解析エラーなどに対する安定であることがわかった (Ishihara et al., 2013; Sugimura et al., 2013)。上記の力推定法では細胞接着面を直線として取り扱っているが、細胞接着面の曲率と力の関係(ラプラス則)を考慮することで、逆問題が未決定から過決定になる。そこで、細胞接着面の曲率を考慮した、新しい力推定法を開発した。細胞接着面の曲率の大きさは組織や発生段階ごとに異なるので、力のベイズ推定法(Ishihara and Sugimura, 2012)と新しい力推定法のそれぞれが、個体発生の異なる局面の力学制御の解明に貢献できると期待される。
細胞集団の協調的振る舞いが引き起こすパターン形成として、ショウジョウバエの上皮組織の細胞競合に注目した。これまでのタイムラプス画像データ(Ohsawa et al., 2012)と比較して、より定量性の高いデータを得られるようになった。タイムラプス画像データに力推定法を適用し、推定した力の動態と細胞分裂、細胞死、細胞形態変化などの細胞レベルの動態の関係の解明に取り組んだ。
