Outline of Annual Research Achievements

本研究では、がん微小環境シグナルに支えられたがん幹細胞性獲得・維持メカニズムを理解することを目的とした。本年度は、raptor遺伝子欠損マウスを用いてmTOR複合体1の白血病幹細胞の増殖・生存における役割を解析した。その結果、白血病幹細胞維持におけるmTOR複合体1活性への依存度は、ニッチ環境の存在下と非存在下では、大きく異なることを見出した。ニッチ環境の存在しない状態でraptor遺伝子を欠損させ、mTOR複合体1の活性を失わせた状態では、その増殖や生存が著しく障害された。また、移植実験により生体内での評価を行った実験においても、比較的分化形質を示す白血病細胞では、細胞死が誘導され、mTOR複合体1への依存性が高いことが判明した。一方で、白血病幹細胞は生体内でmTOR複合体1非依存的に生存していることが明らかとなった。興味深いことに、これらの細胞は、長期的に生存するものの末梢血中での白血病の顕著な増殖は認められなかった。しかし、raptor遺伝子を導入することによって、mTOR複合体1の活性を復活させることにより、野生型と同様の白血病の発症が見られた。このことから、白血病幹細胞は、微小環境から支持され、mTOR複合体1低下に対する抵抗性を示し未分化性を保つことができると考えられた。本現象は白血病治療後の微小残存病変と類似しており、そのような病態での細胞内mTORシグナルの抑制状態と白血病幹細胞ニッチ双方の重要性が示唆された。これらの研究により、がん幹細胞の治療抵抗性メカニズムを理解する上で重要な知見を得ることができた。
