Outline of Annual Research Achievements

本研究は、固形癌での個体レベルにおける癌幹細胞の性質解明を目指し、癌幹細胞を標的とした癌治療法の可能性を示すことを目標とた。
安定型β-catenin inducibleマウスを作製し、成体マウスにβ-cateninを強制発現させた。β-catenin強制発現後早期にde novo crypt formationが誘導されることが分かった。β-catenin強制発現によりcryptを維持する幹細胞が増生することが示唆された。実際にβ-catenin inducibleマウスを大腸上皮幹細胞マーカーであるLgr5 レポーターマウスと交配することで、Lgr5陽性細胞数が増加することが確認された。一方で成体マウス腸管にdominant negative TCF4を強制発現させると、腸管crypt数が減少することが分かった。腸管幹細胞の維持にはcanonical Wnt pathwayの活性化が必須であることが確認された。β-cateninを強制発現させた腸管上皮には、β-catenin蛋白蓄積のheterogeneityが観察された。核にβ-cateninが蓄積した腸管上皮細胞は、細胞増殖活性が低く、正常腸管幹細胞の性質に類似することが分かった。さらにNotch pathwayの阻害によりslow cyclingな幹細胞様細胞に強い細胞増殖活性が誘導されることを見いだした。ヒト大腸癌においても癌細胞でのβ-catenin蛋白蓄積のheterogeneityが観察され、この安定型β-catenin inducibleマウスは人工癌幹細胞モデルとなりうることが示唆された。本研究により、腸管上皮の分化/増殖には、Wnt pathwayとNotch pathwayの協調的な制御が関与していることが明らかとなり、それらが癌幹細胞に対する治療法開発の標的となりうることが示された。
