Outline of Annual Research Achievements

強直性脊椎炎(AS)感受性ERAP1依存的なHLA-B27提示リガンドの同定に向けた培養細胞株を作製した。テトラサイクリン誘導体(Dox)により遺伝子発現をコントロールすることが可能な細胞株Tet-off HEK293細胞を用い、まずその内在性ERAP1の発現量をshRNAによって低下させた細胞株HEK293/koERAP1細胞を作製した。次いでHEK293/koERAP1細胞にx6 Hisタグを付加したHLA-B*27:05遺伝子を安定に発現するHEK293/koERAP1+B27細胞を樹立した。最後に別途作成したshRNA耐性となるように同義置換を導入したAS感受性ならびに耐性ERAP1遺伝子をDox感受性プロモーターの下流に挿入したプラスミドをHEK293/koERAP1+B27細胞に導入することで、HEK293/koERAP1+B27+rsiERAP1(WT/K528R/Q730E)細胞株を樹立した。本細胞株を用いて各SNP型ERAP1の発現誘導に伴う細胞表面HLA-B27の変化を解析した結果、いずれの場合も細胞表面HLA-B27発現量が低下していたが、AS耐性型である野生型ERAP1の場合がHLA-B27量の低下が最も大きく、AS感受性であるK528RならびにQ730E ERAP1では小さな変化しか認められなかった。これらの結果から、樹立した細胞株がAS感受性ERAP1依存的なHLA-B27リガンドの同定に適していることならびに各SNP型酵素が作り出すHLA-B27リガンドのレパトアが大きく異なっていることが示唆された。
