Outline of Annual Research Achievements

本研究では、ヒト6番染色体のHLA領域における薬剤感受性について検討する。そのために、次世代シーケンサーによるHLA領域の詳細な変異検索をおこなっている。
薬剤は時に、有害な反応(副作用)を示す場合がある。その副作用が重篤な場合には、患者が死に至る例もあることから、その機序解明は目下の急務である。特に重篤な薬剤副作用の発生により薬剤が市場から撤退する事例もあり、個々人の遺伝的背景と薬剤副作用の関連は注目されてきた。近年、ゲノムワイドな遺伝学的関連解析の結果から、特定の薬剤副作用とHLAアレルとに非常に強い関連があることが明らかになってきた。しかしながら、どういった分子(薬剤、薬剤産物とペプチドの複合体が予測される)によって副作用が惹起されるか、かつ関与する免疫応答機序もまったく不明である。
以上を踏まえ、本研究では、HLAアレルと薬剤副作用に関する原因分子の同定、及び発生機序の解明を目的とする。そのため特異的なHLAアレル発現細胞を作製し(HLA-A*33:03)、塩酸チクロピジン投与後、HLAアレルに結合しているペプチド分子(薬剤もしくは薬剤代謝物との複合体)の同定、及びそれを用いた免疫応答機能解析を試みた。同時にコンピュータモデリングをおこない、HLA-A*33:01のペプチド結合領域に相同性を有する薬剤代謝産物の同定をおこなった。
