Outline of Research at the Start

幼少期の運動は脳の老化を減速させる一方で、成人後の身体不活動、座位時間、喫煙、睡眠不足、肥満、低体力は脳の老化の加速と関わる。それでは、これらの身体的健康要因はどのような分子生物学的メカニズムで脳の老化と関わるのだろうか?本研究では、DNAメチル化の役割に着目し、身体的健康が脳の老化を遅らせる分子生物学的メカニズムを調べることを目的とする。具体的には、身体的健康と脳の老化の関係を媒介するエピジェネティック年齢(i.e., HorvathAge、HannumAge、SkinBloodAge、PhenoAge、GrimAge)およびゲノムワイドのDNAメチル化を調べる。

Outline of Annual Research Achievements

身体を健康に保つライフスタイルは、脳老化の減速と関わることが示されている。例えば、習慣的運動に伴う体力の向上は、中高齢期の脳老化の減速と関わる。一方で、過度な座位時間、喫煙、睡眠不足、肥満、高血圧は脳老化の加速と関わる。それでは、これらの身体的健康要因は、どのような分子生物学的メカニズムで脳の老化と関わるのだろうか?本研究では、DNAメチル化の役割に着目し、身体的健康が脳の老化を遅らせる分子生物学的メカニズムを調べることを目的としている。
この目的を達成するために、身体的健康指標(過去・現在の運動習慣、身体活動量、睡眠習慣、喫煙習慣、体組成、肥満度、筋力など)と磁気共鳴画像法を用いて取得した脳画像データ(脳容量、大脳皮質の髄鞘密度、神経突起密度と方向散乱、構造的・機能的領域間結合など)の関係を調べ、それらの関係を媒介するDNAメチル化指標を同定する。
令和5年度には、95名分のDNAメチル化解析を行い、身体的健康指標および脳画像データとDNAメチル化から算出された生物学的年齢の関係を分析した。その結果、身体的に健康な者ほど生物学的年齢が若く、脳の老化が抑えられていることが示された。
