Research Abstract

生体分子を化学修飾する分子プロ－ブは生命現象の制御や解明に不可欠であり、その重要性が認識されている。既に蛋白質を化学修飾する低分子プロ－ブは種々検討されており、細胞内において選択的に標的蛋白をラベル化する分子プロ－ブも開発されている。しかし細胞内でDNAやRNAを配列選択的に化学修飾する方法論は現在のところほとんど報告されていない。近年、RNA研究の進展に伴い様々な機能性RNAの存在が明らかにされており、細胞内でこれらのRNAを選択的に化学修飾できる分子プロ－ブは、その機能解明のツールとして極めて有効であると考えられる。本研究では塩基欠失部位を持つオリゴヌクレオチドと反応性核酸を含む低分子プロ－ブを用いた、DNAあるいはRNAの1塩基に対し、ピンポイントの選択性で化学修飾する方法論の開発を目指した。
昨年度までの検討において、独自に設計した水素結合形成により活性化される反応性塩基とDNAに対して結合親和性を持つ部分として2本鎖DNAのAATT配列を持つマイナーグルーブに結合するプロリンーアルギニンーグリシンーアルギニンープロリン(PRGRP)のペンタペプチドを持つプローブの合成に成功した。さらにえられたプローブを用いて、塩基欠失部位を持つ２本鎖DNAとの反応を検討したところ、塩基欠損部位の向かいのチミンに対して反応することがわかった。
そこで今年度はこの結果に基づきさらにより高い反応性を持つプローブの開発を目指した。前年度の検討からカチオン性の高い部分がDNAに対して結合するのに必要であることがわかったため、PRGRP部位のかわりにペンタアルギニンを導入したプローブを合成した。これらを用いて２本鎖DNAとの反応を検討した結果、中性条件下、24時間後、約６０％の収率で塩基欠損部位の向かいのチミンに対して選択的に反応することがわかった。
