| Research Abstract |
天然物リガンドの重要な標的分子であるGタンパク質共役型受容体(GPCR)に着目し、リガンド未同定のオーファ
ンGPCRの天然リガンド同定と、生理活性を有する天然物リガンドが標的とするGPCRを明らかにすることを目的に研究を推進した。
マウスの臓器からBligh&Dyer法で脂質を抽出し、アセトン分画によってアセトン可溶性画分と不溶性画分に分画した。それら脂質画分を用いて、約30種類のオーファンGPCRの活性化能を検討した。GPCRの活性化能は主に、あらゆるGタンパク質の活性化能を検出可能なTGFalpha shedding assayを利用した。その結果、臓器から抽出した脂質画分に、複数のオーファンGPCRに対する特異的な活性化能があることを見出している。現在、臓器からの抽出条件、各種臓器の検討、GPCR活性化能の検出方法の検討を行っている。本研究により新規の脂質メディエーターが同定され、新しい生理作用の発見が期待される。
また一方、生理活性が報告されているが、受容体が未同定、もしくは報告されているが再現性に乏しい脂質メディエーターが存在する。そこで、それら脂質メディエーターが標的とする受容体のスクリーニングを行った。リガンド活性能は主にTGFalpha Shedding assayを用いた。その結果、少なくとも１つの脂質メディエーターが標的とする受容体の同定に成功し、さらなる詳細な検討を行っている。
