Outline of Annual Research Achievements

本課題では、窒素の特性を生かし、エナミン機構に基づき反応を加速する不斉有機分子触媒を設計、創製すると共に、それらを用いる反応法を開発することを提案した。生物活性分子や生命機能解明に用いる機能性分子の開発のためには、生体分子と相互作用するための構造単位として、酸素や窒素を含む官能基、ヘテロ環等を含む多様な分子を合成することが必要である。本研究では、多様な置換基や官能基を有する一連の化合物を、穏和な条件下、簡単に、短工程で合成する方法を開発することを検討した。特に、アミンを触媒とし、反応系中でエナミンを生成することを鍵とし結合生成反応を加速する反応およびその触媒の開発、および、高活性触媒を得るための戦略の開発について検討した。
1.エノンとイサチンの不斉ヘテロディールスアルダー反応の開発
テトラヒドロピラノン構造およびスピロオキシインドール構造は、生物活性を示す分子やその合成中間体によく見られる構造であり、これらを組み合わせた分子骨格上に、種々の置換基や官能基を配置した分子群は、生物活性等を示す分子を探索、開発する上で有用であると期待される。本課題実施1年目に開発した、反応系中、エノンからエナミンを生成させ、これをジエン成分とし、イサチンとの反応により、高ジアステレオおよび高エナンチオ選択的にテトラヒドロピラノン環を構築することにより、スピロオキシインドール誘導体を合成する方法について、反応進行および立体選択性の発現に重要である因子を明らかにする検討を行なった。
2.エノンを原料とする3-アシルピロール誘導体の合成反応の開発
3-アシルピロールも生物活性を示す分子やその合成中間体によく見られる構造である。エノンへのアミン誘導体のアザマイケル反応によるC-N結合生成、引き続くC-C結合生成、そして酸化反により、多置換3-アシルピロール誘導体をワンポットで合成する反応を開発した。
